The Amazing Spider-Man #6 (November 1963) is the debut of Dr. Curtis Connors as the Lizard — one of the most enduringly sympathetic figures in Spider-Man's rogues' gallery, a war veteran and scientist who loses control of his own humanity through desperate self-experimentation. The character's Jekyll-and-Hyde dynamic gave Lee and Ditko a template for the morally complex, tragedy-driven villain that would become a hallmark of the Spider-Man mythos, distinct from the purely malevolent antagonists common to superhero comics of the era. The issue also marks the first appearances of Martha and Billy Connors, establishing the family unit that has given the character emotional stakes across six decades of stories. Its influence extended well beyond the printed page: the Lizard's origin powered the primary villain arc of the 2012 Sony film The Amazing Spider-Man, and Connors (in human form) has appeared across multiple live-action, animated, and video-game adaptations.

Written by Stan Lee and penciled, inked, and cover-drawn entirely by Steve Ditko, the issue arrived in November 1963 as part of the rapid-fire world-building that characterized the Lee–Ditko run on the title. Lee and Ditko were at this stage operating under the 'Marvel Method,' in which the artist worked from a rough plot outline and Lee scripted the dialogue over finished pages. Letterer Artie Simek and colorist Stan Goldberg rounded out the production team. The issue's letters column, still buzzing over the young series' early success, reflects the collaborative, fan-facing culture Lee was actively cultivating at Marvel during this period.
