The main story features Sir Spot the lion-hearted leopard and Droopy the dog discovering a pirate's treasure chest. When Droopy struggles to open the heavy chest, he hits it with his head, causing it to spring open and revealing gold coins and a treasure map. Droopy then travels to college where professors study a brain, planning to sell the treasure once the professors finish their research. A group of crooks pursues Droopy to steal the gold bullion, leading to a chase involving a bloodhound detective, various animal characters, and slapstick mishaps including the loss of Droopy's diaper and the destruction of furniture and a movie camera before the crooks are apprehended at a cliff.
